Abstract
Nowadays, infections caused by Pseudomonas aeruginosa and fighting against them are a priority of clinical medicine that is associated with increased resistance to antibiotics of this microorganism.
Therefore, interest in phagotherapy in the treatment of diseases caused by this opportunistic pathogen is increasing not only in Georgia, but worldwide.
Pharmacological studies are the previous preparatory stage for the introduction of medicines, including phagic drugs, in the clinic, which is of great importance in terms of their effectiveness and safety. Nowadays, the rational use of phagic therapy requires a detailed description of the candidate phages and the study of pharmacokinetic and pharmacodynamic characteristics.
The objective of this study was P.Aeruginosa specific phage called PNMx, which was separated from the Mtkvari water in 2018. For the above mentioned phage, as the proposed candidate in the experimental-therapeutic preparation we have studied the different biological features: lysine spectrum, multiplication dynamics, the frequency of generation and stability of phagospecific mutants in the thoracic area, for which we have used semi-purified phagic drug “phagolysate”. the same drug was used to study the pharmacokinetics and pharmacodynamics on the mouse model, and high purity phagine preparation (CsCl gradient cleansed) was used for studying the phage virion morphology and DNA restricting profile. The study used standard bacteriological and immunologic methods for research on phages.
